Quickest way to rule out trans! Try to power brake it. This will put enough of a load on the engine to rule out the trans problem. If it still has a miss when power braking, have someone put the truck in reverse and power brake a load on it while you disconnect and reconnect each fuel injector harness. This will pinpoint the cylinder with the miss by not changing how the engine runs on the dead cylinder. This is the cheapest and easiest way without a scanner or a hard code set.
If you want to try what was suggested earlier to force a check engine light something else to consider which might help force the light and which is how i got mine to come on is to hook up to a trailer with something like a lawnmower or just empty if its a big trailer so you have a 1000 or so pounds to put extra load on the engine and then get out on the highway and floor it, i just pulled out and mashed the gas and it shook and rattled like it was gonna fall apart but i got the code. I had a similar problem and it was with my coils after that i just drove to autozone to get the code checked. Luckily i replaced the coil that popped the code and it fixed my problem but just replacing them all might be the way to go. It cant hurt to try it as it will only cost you a few dollars in gas, autozone will scan the codes for free.
