Hi and welcome to F150online. Congrats on your new truck,
As for which tuner is the best, do you mean the person ot the equipment?
The tuning device is just the delivery equipment for the "tune" which is written by a person.
Personally, I went to Troyer Performance, had a chat with Mike Troyer and discussed my tuning requirements.
He went over with me what I wanted performance wise and also my budget and servicing intervals.
I ended up going with an Xcal2, pulley and intake package from him with 3 custom tunes.
My HD runs awsome now and I couldn't be more pleased.
However, there are plenty of other tuners out there but, to me, you can't beat a personal recommendation.

Yes, I did dyno once but the results seemed off on quite a few cars there including mine on the day.
Just to point out, all dyno's read differently from each other so saying that you get 400hp with this and that is not always a true indication of output.
A Dynojet reads differenlt to a Mustang dyno and then you have how the correction factors are applied.
However, using a performance calculator http://www.wallaceracing.com/et-hp-mph.php and pluging in the factory figure of 340hp for the HD when stock, gives a quarter mile time of 14.82 seconds.
I ran a best of 14.92 stock so that's pretty close to the real result.
Using the same calculator, entering my new best time of 13.50 seconds with the same weight gives a computed hp figure of 450 hp.
So, what can be deduced from that. You could say that my modifications have been worth approximately 100hp over stock.
