Fault codes

I hooked up my OBD2 to my daughters 01 Escape because the CEL came on today. I am getting a P0301 misfire in cyl. 1. What is the first thing I should go after to fix my problem? I do have mechanical ability but not much experiance with engine repair/diagnosis. I know there are people on here that can help, hopefully without being smartarses.

Alittle over 111.xxx on the clock. Yes it is a V6. And yes I think I'll change out the coils and plugs on cyl.1,2,3. They are under the intake manifold and so far it isn't fun to get apart. So might as well change them all why I'm under there.
