You can remove the main driveshaft, and move the truck using the front half of the 4x4 system.
You'll lose transmission fluid through the rear of the transmission, and I don't think the front half of the drivetrain was designed to handle 100% of the engine's power.

What is wrong with your rear diff that you want to disconnect it?
Sure you could drive in 4x4 without the rear driveshaft installed (I don't personally know if the transmission would leak fluid from the tailshaft w/o the drive shaft).
BUT if there is something wrong in the rear axle assembly it will still destroy itself regardless of whether or not it's connected to the transmission since it will still spin while the truck is moving
