How did you check the spark, and how do you know that cylinder isn't firing?

I know its not fireing cause its the only cylinder that doesnt make a difference when i pull the boot off the plug. all the others make a difference in how it runs when i pull the spark plug boot off. i checked the spark by getting another plug and just holding it up to something so i can see if it arked.

First things first. Tell us what engine you have
Then check your compression in all cylinders, not just the allegedly bad one. If you have a mechanical issue, you need to find it and fix it before you screw around with fuel and ignition
