Cylinder Misfire, Etc.

I installed the Edge Evolution CTS and back up camera yesterday on my 2001 F150 4X4 Lariat SC Flareside. I did this because my check engine light was coming on all the time before my lift was installed. I figured I could use the programmer and DTC info. The truck has been running OK but this AM I ran an errand and the truck ran very rough when I was leaving the auto parts store. Go figure! The following codes came up: P1000-on board diagnostic OBD system readiness test not complete; P1405-plugged or open upstream HI side DPFE sensor hose; and, P0301-cylinder 1 misfire detected.

I'm no mechanic but I am handy and need to save $$ if possible. My 5.4 L V8 has 178,400 plus miles on it but the truck has been well maintained by me and its first owner. I want to keep this truck and the new suspension, wheels and tires look great. I want to keep it for show and tow. Should I go to the mechanic or can I fix these issues? I have the two volume Ford shop manuals for my truck.
